This hands-on laboratory course introduces undergraduate students to basic techniques used in molecular diagnostics. Students develop practical skills in genomic DNA extraction from biological samples, measurement of DNA concentration and assessment of DNA quality, and separation of nucleic acids by gel electrophoresis. The course introduces polymerase chain reaction (PCR) and its diagnostic applications, including quantitative real-time PCR (qPCR), reverse transcription PCR (RT-PCR), multiplex PCR, amplification refractory mutation system PCR (ARMS-PCR), and PCR?restriction fragment length polymorphism (PCR-RFLP) analysis. Students also learn the principles of DNA sequencing and interpretation of sequencing results. Throughout the course, emphasis is placed on laboratory safety, accurate pipetting, quality control, experimental design, data analysis, and critical interpretation of results. The course provides a foundation for further study and practical work in molecular biology, genetics, and biotechnology.
